"Best" option I can think of so far is to go for AA and pay for a MCE upgrade.
I also have a decent number of Amex MR points that I could convert and use for a points upgrade (albeit flight would likely be booked in lowest economy fare, which may exclude me from points upgrades)

Points upgrades are rarely, if ever, a good deal.
Are you looking simply to upgrade to a main cabin extra type product? That is usually a fixed fee. Confirmed business class upgrade costs will rarely be cheaper than buying a business class ticket to begin with.
You dont say your origin and destination, but if miles are not important to you, you should look at carriers that provide lower-cost business class options, which range in quality, including:
- Norse Atlantic "Premium" seats
- Icelandair "Saga Premium"
- Condor - both premium and business
- La Compagnie- all business class, frequent sales
