The Flagship First Check-In access rules are fairly clear:

You can use Flagship First Check-In if you’re flying on American in Flagship First or Flagship Business Plus on a qualifying international or transcontinental flight.

You can use Flagship First Check-In if you’re flying on qualifying international flights operated by American (regardless of cabin) with:

AAdvantage Executive Platinum status
AAdvantage Platinum Pro status

ConciergeKey members

Non-AAdvantage oneworld Emerald

You can use Flagship First Check-In if you’re flying on any flight operated by American (regardless of cabin).*
*Excludes Mileage Plan MVP Gold 75K

Five Star Service customers
Executive Platinum and Platinum Pro AAdvantage members can use it, Alaska MVP Gold 75K members cannot.
