Yes and yes. You have a connection (ADD) on your outbound flight and you can have another connection (or stopover) on your inbound/return flight. Either IST or CAI would work as a connection or stopover (because both IST and CAI are considered by ANA to be in the "same" area as your destination), and returning to either DTW or JFK is fine because they're both in North America (you can even return to Canada if you can't find a return flight to US).
