With the introduction of premium select on transcon flights, it looks like Delta has updated it's
rules for upgrades with regional upgrade certificates:
- Flights with domestic Delta One and domestic Delta Premium Select:
- If you purchase Main Cabin or Delta Comfort+ and then apply your Global Upgrade Certificate, you will be upgraded to Delta Premium Select upon request and then will be added to the waitlist for Delta One and eligible to clear starting 24 hours prior to departure. From the reading of this, it looks like RUC's are no longer eligible to be used for upgrades to Delta One on transcontinental flights
It's not exactly new as that text has been there since May 2023. There are detailed T&C's at the bottom of that page for RUCs --

The Certificate may be applied toward a one-cabin upgrade. If a flight has three or more cabins of service, the Certificate will permit an upgrade of only one cabin beyond the cabin selected for the purchased ticket; additional Certificates cannot be applied to the same ticket to upgrade more than one cabin beyond the purchased cabin of service. Delta representatives must book in “RX/OX/OL" class and; travel agencies must book in "R/O" class."
