Yup! I do it all the time. Just tell the return agent you're doing a back-to-back rental and that you'd like to keep the same car. They'll likely tell you to hang onto the keys and go to customer service to have the car information added to your new rental reservation.
I do it all the time, too. Make sure the tank is full in the unlikely event they require it back. Particularly at an airport location, make sure to keep the keys with you.
