If you value walkability I would go with Hotel Vancouver as you are much closer to the centre. You can literally head in any direction and explore a different part of downtown. Waterfront is good if you are focussed on the Canada Place/Gastown area, or if you don't mind the extra 10+ minutes to reach the southern parts of downtown.
