From experience this hasn't been possible directly through booking however I have contacted hotels I have stayed in numerous times via email and always found them to be accommodating. Rooms are generally priced on 2 people but best to contact the hotel. I know friends who did this in Las Vegas without telling the hotel and some guests had a bit of a job trying to explain this when checking in.
