Call IB and explain that your wife has covid. Iberia should allow at least free rebooking, if not full refund. You may have to submit evidence of Covid infection though. Ib has no interest in your wife traveling on their flight(s) with Covid so they will usually cooperate.

If everything fails with IB, you contact your travel insurance for reimbursement.
