The '5 night' rule perplexes me as well from a car rental perspective. It worked out in my case because the cheapest flights were 11/1 to 17/1 (i.e. 6 nights), so I figured paying a few quid extra was worth it for the peace of mind, and the BA website seems to 'agree' with my selection.
I don't have an answer as to why the rental from 11/1 to 16/1 doesn't count as 5 nights, maybe someone else might.
EDIT: from crazy8534's response below, it seems like this is just another BA IT gremlin?

The absence of the "double tier points" tagline on the booking doesn't really relate to its eligibility.
If you have 5 "over midnight" car nights booked or 5 hotel nights with a UK start then you will get the double TPs.
The second booking in the quoted post would be eligible.
