Do it when you pay. It is available at the last step at payment when you scroll down. Unless you are booking a holiday that the automated system doesn't accept for a deposit like an open jaw or complex booking.
Using Avios to part pay towards a holiday can only be done at initial booking stage. Remaining balances of holidays secured with a deposit cannot be paid by using Avios.
I have to say that it looked like a spectacularly poor use of avios so I ignored it.
