Without any authority to answer this question, I would tell you that if it were me, I'd feel ok making the OTA booking and reaching out to the hotel to let them know. The property shares your goal in that they would rather keep you in the same room than move you twice. So assuming you're not seeking an upgrade, there's no reason why they wouldn't make a note and do their best to keep you in the room (there's always a small chance something goes wrong and they can't for any reason).
