The proper way is to enter using the regular lane to have yourself admitted by an officer using the new O1 visa first and then update your new visa at an enrollment center for future GE usage.

By the book, all updates are to be done at the Enrollment Center (EC) only but it doesn't hurt to ask the officer to see if the Enrollment On Arrival (EOA) officer can assist with updating your profile.
If the EOA is not backed up, and you're polite about it, they are capable of doing the update since it's the same system that the EC uses. But if they say no, just don't push it and hurry to the EC or wait until you're at the airport again. EOA has no obligation to do the update.
