Most ICs require either a Friday/Saturday or Saturday/Sunday stay, but at some Middle East hotels the free weekend night is for Thursday/Friday or Friday/Saturday stays. From Ambassador T&Cs:
'Weekend' is determined as follows: Friday, Saturday and Sunday, except in the Middle East where hotels may define the weekend as Thursday, Friday and Saturday
So, it depends where you plan to use the certificate. At the majority of ICs what you are suggesting would not be possible. The simplest way to check is to initiate an Ambassador free night certificate booking to see whether it is available on the dates you want.
