I've never booked on a Virtuoso rate before, but I've booked through similar TA channels, such as Travel Leaders, and have never been told I couldn't redeem my points at the hotel. I'm sure there's something in the T&Cs about only being able to redeem points against a stay eligible to earn points. But, I've never had a stay through a TA like that not be eligible to earn points either, so I don't see why you can't.
