Not likely, especially on LEVEL.
PE is a tiny cabin whilst Y is massive. The only incentive airlines have to provide discounted upgrades is when one cabin is nearly full and they might have to upgrade for free - so check if somebody will pay for that upgrade.
That mostly works best for booking PE and trying to upgrade to J later. But that's not relevant with LEVEL.
If you want 4 seats in PE, you will have to book 2 using Avios and the other 2 at the standard cash price. Note that booking the 2 award seats first might cause the cash fare to increase.
