Prior to check in on a single PNR you will not be separated but may miss an upgrade if at the top of the list and only one seat is available.
After check-in if a single seat opens and you are at the top of the list, the PNR will be divided and one will be upgraded. The other may or not be upgraded later. The only way to avoid being separated is to delay checking-in early and when you decide to check in withdraw the waitlist request. Of course that means you will miss the opportunity for last minute upgrades.
Note once you get closer to departure you will be able to see your place on the waitlist. 3 days out (and if you play with your device date once within 7 days, setting the device to the day of the flight).
Being in PremPlus helps your odds of upgrade, some some status would help further.
