I've had TSUs not get deducted immediately after application on both points and cash reservations. Most if not all times it got deducted eventually.
In general, for peace of mind, after applying a TSU I tend to reach out to the hotel directly, rather than Hyatt corporate, to confirm that I've been blocked into a suite and everything else looks good (and make any special requests). I do this regardless of whether the TSU has been pulled and/or the suite is showing up on my reservation.
