I certainly agree that it can be done during the booking process providing U space is available but I too am unaware if an existing itinerary can be upgraded with miles even if an upfare is required.

Personally, I wouldn't upgrade with miles but you do bring up an interesting point. It isn't one that I had considered before. Economically, though, it doesn't make sense in many cases since the lowest underlying fare eligible for a mileage upgrade is more than the sell-up fare into F.
