I doubt it. If you had booked the nonstop originally with Avios and then had to be re-accommodated on connecting flights, you would not be expected to pay extra Avios.

might be entitled to a refund of the second passenger-facility charge (PFC) since you are no longer connecting, but that would only be a few dollars.
