1) You need to be absolutely sure which terminals your flights will be coming into and going out of before anyone can answer this
2) Double and triple check that Air Nostrum will allow you to through-check your bags to the final destination. If they use the same IT systems as Iberia they are very clunky and often baggage through-checking creates issues. So don't just assume this will happen, be absolutely sure that it will.

Provided that you can through-check and provided that you're doing T4 to T4S and provided that the first flight arrives on time, you should be OK. Immigration lines at Madrid are IMO about 40 mins maximum for non-EU, depending on which other flights have arrived at around the same time as yours. So that will give you time.

If you're on one single booking reference and you miss the connection, it's the airline's responsibility to get you to your final destination, so I guess it's also a factor how terribly you'd be affected if you got to HKG a day late.
