We did call JAL in Japan and they were not willing to touch the ticket. Perhaps might have better luck with JAL in USA. I wonder if showing up at JAL ticket desk at the airport would help. I'm also going to try with Amex again and demanding that they keep JAL on the phone until I have resolution. The Amex agent's comment that "I need to respect JAL agent's time" from earlier today really infuriates me lol.
I wouldn't waste time at the airport, they can't really do anything outside the scope of the flight they are working that day. They couldn't/wouldn't even check and change a seat assignment for me on a future flight within the same itinerary.
JAL USA call center supervisor is the best bet now while you're waiting for Amex tomorrow.
