Aren't there also capacity-controlled premiere upgrade seats that can be scored at booking time for full Y fares? i.e., automatic business class seat at booking when booking full Y?
My understanding is that if you miss out on the opportunity at booking time, you have to wait much later, until the regular upgrade queue even if the capacity becomes available in the meantime.
Is there any way to upgrade into business if using DTS and booking a city pairs Y fare?

Sorry, I overlooked your careful usage of "preferred" seating.
Different federal agencies have different rules on required booking with a specific travel agent, such as CWT SATO.
When a reservation is under control of a travel agent, UA will not allow substantive changes to the reservation by its own agents.
I am not sure if seat assignments, which are not supposed to have a charge, fall into that category.
I would call uo UA, and if that doesn't work, call up CWT SATO.
I have found that United is much less restrictive than other airlines. Alaska won't touch a booking made by SATO. United treats my city pair tickets as Y class, allowing me to make all sorts of changes within their app.
