Is it possible to book this? I know you can return from a different departure point on a 2-4-1, something in the back of my mind is flagging that the flights have to be a similar distance to qualify , or have i got this wrong and the above is ok?

As someone else has said, the distance between your destination to (FRA) and from (JFK) needs to be shorter than the distance from London to each of the destinations. So in this case, it definitely will not work.
Something like LHR-JFK and then MIA-LHR will work since the distance from JFK-MIA is shorter than the other 2 legs.
There are some very creative ways of using this rule which I have heard of but in your case, unfortunately the answer will be no.
