Yes you can do that, but it kind of defeats the purpose really. The idea of Twillight check-in is to avoid the usual morning crush. You may as well check everything in on the morning and at least that way you only have one excursion to check-in. If you are in Club then you automatically get Premium Gatwick so you don't need to pay for it separately.
