I mean, of course Amex is going to say it is effective immediately. If the purchase isn't time sensitive, and I can't imagine a purchase of even more space seats is, I'd just wait until the next day out of an abundance of caution. There seems to be no rhyme or reason for the data points that say it works the day of selection or not.
