Yes, the KTN field in a previously made flight reservation can be updated with a new KTN upon the traveler getting a new KTN number. It is best to try to update the KTN number with the new number at least 72 hours before flight departure, but it can even be done on the day of check-in with ticketed bookings made even months and months ago. If your booking is connected with a DL SkyMiles number, also update your KTN in the Delta SkyMiles profile.

I do the updating of the reservation’s KTN online on Delta.com, but it can also be done by calling the airline. I have even done it at the check-in counter.
