Yes, an ESTA is required for transiting the US. The US does not have airside transit, so he must be fully admissible to the US, clear immigration and customs in the US, and then recheck his bag and go through TSA, etc. to continue to his onward flight.

The ESTA form has a field to indicate that he is only transiting, and an address in the US is not required.
