If the TSU would go to waste then go ahead and use it. But if there's a chance you might use the TSU later then save it, not worth burning for one night simply hoping to get a further upgrade. To answer that question though, it isn't unheard of. I've read a number of DPs where a globalist with TSU was further upgraded. It ranges from impossible to a solid maybe (never likely) and is property dependent.
I've never personally done this, but it's apparently possible to negotiate at check in time if you have not applied it yet. My friend likes to do this.
Say he gets to a property and sees that both standard and premium suites are available. He gets the comp standard suite upgrade. He asks about the premium suite, and they say it's not complimentary but they quote him a price. Then he says, what if I apply the upgrade award? Often they will agree to the upgrade to the premium suite in exchange for applying the upgrade award.
