It depends. Let's assume that DL doesn't make mistakes in crediting the flights. (YMMV)

If you call DL to change the ticket and pay the fare difference, it will be treated exactly as if you had purchased the higher fare class originally.

If you pay with money or miles to upgrade a single segment on the app, you're really just paying a fee to sit in a different cabin (and get that cabin's service and other perks). You still have a coach ticket.

If you use a GUC/RUC or take a mileage upgrade award, you earn based on the original ticket purchase.
