It's not just a matter of the mandatory quarantine. Coming from the US, you won't be admitted to Japan at all unless you're a Japanese national or returning permanent resident, unless you get an "exceptional circumstance" visa from the Japanese embassy, which they're certainly not going to issue for an overnight transit passenger.
