Technically, no. The Terms of Conditions of the Hilton Honors program state:
"
To earn Hilton Honors Points or credit toward tier status for hotel stays, a Member must be a registered, paying and staying guest at a participating hotel within the Hilton Portfolio and meet all of the conditions described herein."

In practice, a lot of hotels will have no problem issuing points in this scenario because the elite-level member is staying in the room. But, you really won't know if she'll get the points/stay credit until after the stay. So, it's a bit of a gamble.
And to make matters more murky, you can't always trust a Front Desk Representative. That person might tell you it's fine without knowing that the Night Auditor will be running a report that identifies mismatches between the guest name and the credit card name in order to deny points. (Not every hotel will check. I worked at a hotel where we absolutely did not care and another that did everything to minimize costs like providing frequent guest points.)
Unless it's a huge burden, I'd strongly recommend letting your wife pay the bill.
