I believe the mile transfer is from a non-Delta source into the wife's account in order to have the necessary number of miles for the last ticket. Assuming my read is right, I don't see how doing a call prior to doing the transfer will be of any use.
Putting that to the side, the OP should do a dummy booking for the 4 tickets to make sure that there are indeed that many award tickets at the award level he is targeting before doing the transfer. The dilemma, of course, is that there is no guarantee that the same number of tickets will be available once the OP has done the mile re-positioning.
