Where to go: Montevideo, Asuncion, La Paz or Santa Cruz?

I am looking to go to a unique place on AA--- considering I'm never on AA, this is almost a once-in-a-lifetime type thing. My key criteria is someplace I haven't been and can't easily get to later. The places in the title fit the bill.
Background: I have been to EZE, GIG, LIM/Cuzco, BOG, UIO, and CCS, and I can easily return, or go to SCL or GRU with CO/NW/DL/COPA. I also have miles I can conceivably use to get to Santa Cruz or La Paz, although it would be an all day thing. I suppose I could conceivably go to MVD by ferry or flight out of EZE, but that would incur added cost and travel time.
Request: advice on which one to visit, and a sense of how interesting or worth the trip the spot would be. ASU, for example, is probably a place I would never ever go to otherwise, but I wonder if the visit is worth the effort.
Constraints: I will fly AA-- but availability seems pretty open and connection times are good. I was looking to do like a 4 day weekend, and probably wouldn't venture far out of the city.
I'm asking this way since there are very very few posts on these less-visited South American cities.
FWIW, I'm fully bilingual English-Spanish and have ventured around Latin America on my own.
