First, you need to transfer those miles to your existing Miles&Smiles account. Then you would be able to buy award tickets or upgrade flights with those miles but those miles won't affect your tier unless you convert them to status miles.
In addition, if you have issues transferring point from AMEX to M&S, I would suggest that to contact the AMEX help desk.
