No actual experience here, but Southwest says you can use a LUV Voucher for "passenger travel" at any Southwest "ticket counter location." Whether a BS upgrade qualifies as the former and/or a departure gate as the latter isn't totally clear. I'd suggest you try at the main counter first (baggage check-in, full service if available).
