I want to fly JFK > SIN, with a long stopover in FRA (I'm based in Europe). I understand there is sometimes more availability for JFK > SIN than FRA > SIN.

Date A: award space for JFK > SIN, only waitlist for JFK > FRA and FRA > SIN.
Date B: award space for JFK > SIN, only waitlist for JFK > FRA and FRA > SIN.
In the above scenario, if I wanted to fly JFK > FRA on Date A, and FRA > SIN on Date B - would I be able to confirm the award straight away without waitlisting?
Did you mistype your example? As written I don't think there's any reason to expect you'd be able to confirm an award since neither date has FRA-SIN availability.
Perhaps the better example would be if date A shows JFK-SIN availability and date B shows JFK-SIN availability, but neither one shows independent JFK-FRA and FRA-SIN availability (which, other than last-minute bookings, is a fairly common outcome in my experience) then can you book the JFK-FRA from date A and the FRA-SIN from date B (technically date B+1)? I suspect not, because married segments are based on connections, and connections on international flights are defined as being no more than 24 hours' stop. But it could still be worth calling if you found such a situation.
