I carried the bag with me on tours in Cambodia, Minsk, Burma and Laos. Sometimes I leave it in the car, but I only do so when I have a driver who remains with the car. Never had any issues so far. Though I generally try not to leave anything I can't be without behind in the car. Basically, passport, wallet and phone; everything else is easily replaceable. I also ask the guide if it's safe to leave a bag in the car and I generally trust their advice.

My general advice: I would definitely not trudge around with a roller bag. I'd put all my least important (replaceable) and heaviest stuff in there and just leave it in the trunk. Then I'd put all the important stuff (passport, wallet/cash, phone, camera, tablet, medication) in the backpack (keeping it as light as possible) and carry it with me everywhere, like a purse.

My one bag, for reference.
I must say that is impressive for a multi-stop trip! That said, I may also explore left luggage facilities, since that seems to be a service offered by larger airports in SE Asia, especially if I am transiting back through those airports.
