I pay directly from my bank account to AMEX. The day after I schedule the payment, AMEX has a message on my account stating that "Your last payment of $xx.xx was received on (insert date) and is being processed. Your balance will be updated in 24-36 hours."

Normally, the payment is applied on that same day after the payment is originally scheduled and the balance updated. So 24 hours, IME.
