They definitely can - I've been using it regularly for about 18 months. There is (usually, not always) someone waiting at the exit of the Flagship check-in area who will take you to the front of the TSA Pre line, so skipping anyone (non-Flagship) who is already queuing. I find this a little bit weird, since typically disabled passengers use this line and you're placed in front of them.
