Skybus (which operates the bus from MEL to the CBD) runs a few buses from the CBD to AVV (to meet the handful of Jetstar flights that arrive there).

Or do you mean you want to go from MEL airport to AVV airport (sorry don't know what is "Downtown Airport)? In what case, it would be possible to take the Skybus from MEL to the CBD and the Skybus from the CBD to AVV, if you time it right. They both stop at Southern Cross.
