Yes, the RPU would be considered used if it clears on YYZ-IAD, regardless of the rest of the trip. However, you can call UA and ask them to apply it to the legs you select.
Your chances of clearing on LAX-LIH are virtually nil; the front cabin is full, and you're extremely unlikely to be first in line in case of a misconnect even with an RPU. Your chances on IAD-LAX aren't much better; it's currently booked 17/20 with only one upgrade booked so far. Your CPU chance is essentially zero, and even an RPU is no guarantee -- there's a good chance they sell three more F seats, and even if they don't, you'll be behind GS as well as any 1Ks on instruments.
