Unless something has changed recently, I believe Amex doesn't check ChexSystems for prepaid accounts.
On the other hand they do blacklist people for prepaid and credit cards (separate blacklists) and in the past they have taken up to a couple/few months to show a prepaid account closed after people initiate the closing process.
