If you do the booking on JAL's website, you can see if a confirmed upgrade is available at the time of booking or if you will be waitlisted for an upgrade.
If you are on the waitlist, you will need to have the miles in your account when it clears. It is normally an automatic process for the upgrade to clear, no miles no upgrade.
