There is no minimum time that you have to be in the third country. But normally on this forum, we recommend that you actually go through Immigration and Customs (in your example, that would be enter into Hong Kong formally). Then go to the airline and check-in for your return flight, after which you'd go through HK Exit Immigration and security. HK doesn't stamp the passport anymore, but an electronic record is kept.
For Chinese TWOV purposes, I suppose it is theoretically possible to stay in the transit zone and not go through HK Immigration, but it would depend on whether your return flight airline can issue you a boarding pass without having to get to the regular check-in desks. I'm not sure we have enough data points to know how the Chinese would consider your return (2nd) TWOV if there is no record that you entered and exiting HK. Just be cognizant that you might be in guinea-pig territory if you try this gambit.
Keep in mind that flights to/from Shanghai-Hong Kong can be delayed, so it would be smart to place plenty of time between them. Two hours (per your example) is not enough time to provide enough contingency cushion.
