When you are logged into delta.com, you can check upgrade availability to Comfort+ to see if it is currently available for the flight(s) you are looking at. IME, as long as it says it is available, I get upgraded to Comfort+ immediately after booking.
ETA: I always check the seat map before booking as well, to ensure there is a non-middle Comfort+ seat.
