If it's a pure Avios booking that has flexibility you may change that booking as often as you like under the rules that govern changes to Avios bookings. One joker is all that you require for a PNR so if the new leg you want has no availability then you may attempt to apply the joker to that leg too. Of course, applying a joker requires the appropriate underlying fare class to be available and that others haven't applied the joker (and used all the space) before you.
If it's a revenue ticket that has been upgraded using a joker and Avios then it's a little more complicated. Your ability to change the ticket will be based on the conditions of your underlying revenue ticket.
I'm not sure what you mean about "once you're booked into a revenue bucket"?
