It depends on the hotel, but room/suite type, rate paid, past stay history/spend, booking channel, etc.. do come into play.

I am friendly with a bar tender at a favorite hotel, who once noticed my room number when I signed the bill, and knew that I was in a certain type of suite on the club level.

At a true luxury hotel, though, at least in theory it should not affect the type of service one receives.
