If yes, will they just charge me the equivalent avios for 1 ticket as the whole ticket is a 2-4-1.

Why should this involve a cancel and rebook? You're entitled to change an award booking. So if WT+ award seats become available later, you would just change the current WT award booking to a WT+ award booking. The cost is the additional Avios, the additional TFC plus the change fee. I'm pretty confident that is the answer every time someone comes up with a question about changing to a different cabin on the same flight on an award booking.

The only thing I am not so confident about is whether the 2-4-1 makes a difference. But I can't immediately see any reason why it should. You can change a 2-4-1 award booking just like any other award booking, as far as I know.

Cancel and rebook comes into play if you're trying to change an award booking to a different route in a different zone. Such a change is not allowed on an award booking, so the only way of securing that is to cancel the entire booking and start again. But that's not what's happening here.
