This depends on the type of operation. Scheduled cargo operators who have full infrastructure at both ends will probably run it with just the 2 pilots. Ad-hoc operators will add a Loadmaster and possibly an Engineer.
I run a B727 freighter on ad-hoc charter operations in the Middle East and Africa and we staff our flights with a standard crew of 5 - Captain, FO, FE, Loadmaster and Engineer. If it is a same day turnaround to an airport with reasonable support facilities, the Engineer sometimes stays home after signing out the daily check. The other 4 will almost always fly - but there are circumstances where we might not send the Loadmaster (for example if transporting valuables or livestock where we would need the jumpseats for those attendants).

In the flight deck diagram on
Boeing's website, there's a seat designated "First observer" - could you explain what role that person would have? Would that be a crew member or passenger/courier?
Also on that diagram are 4 seats labeled "Supernumerary seats" - would those be similar to the jump seats for FA's, or more substantial?
