You most certainly CAN combine a Joker with a GUF1/GUF2! You need to phone BAEC's GGL line. They will use the Joker to open up availability in the next higher class and use the GUF to get into it. You 'pay' the avios for the lower class. I just did it today! Used avios for World Traveller, then a Joker+GUF1 to put our daughter in Word Traveller Plus from TLV-LHR-JFK-LHR-TLV. I think that it only works with straight avios bookings though, not cash or part cash/part avios.
If you do a cash booking you can use a joker to open up reward space in the higher cabin and then use the GUF to actually do the upgrade.
If you are upgrading a reward booking and you apply the GUF at the time of making the booking you only need to use the joker to open up the space in the cabin you are upgrading to, reward space in the cabin you are upgrading from is irrelevant. Obviously this needs to be done on the phone.
